Product Specific Information
Description:
The 4S.B3 monoclonal antibody reacts with interferon-gamma (IFN-γ), a 17 kDa cytokine produced by activated T and NK cells. IFN-γ exhibits antiviral and antiparasitic activity and modulates immune responses by interacting with high-affinity IFN-γ receptors on various cell types. Glycosylation does not affect its biological activity.
Applications Reported:
Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Prediluted and validated for intracellular staining using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended use at 5 µL (0.5 µg) per test in 100 µL final volume. Cell numbers can range from 10⁵–10⁸ cells/test.
Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) is a tandem dye emitting at 797 nm, designed for UV (355 nm) laser-equipped cytometers.
Use Super Bright Complete Staining Buffer (Product #SB-4401) or Brilliant Stain Buffer (Product #00-4409-75) to minimize non-specific polymer interactions.
Light Sensitivity:
Protect vial and stained samples from light to avoid photo-induced oxidation.
Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333).
BUV805 is compatible with short-term methanol-based fixation (<1 hour).

Target Information
IFN gamma (Interferon gamma, Type II interferon):
A macrophage activation factor produced by T-lymphocytes and NK cells in response to antigens and cytokines.
It functions as a homodimer (~45 kDa) and induces antiviral, antiproliferative, and immunomodulatory effects, including MHC induction and macrophage activation.
Human IFN-γ is species-specific and does not cross-react with mouse IFN-γ.
Mutations in the IFN-γ gene are associated with aplastic anemia.
