Product Specific Information

The RPA-T8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8α/β) or homodimer (CD8α/α). CD8a is found on thymocytes, mature T cells, and NK cells. It binds to MHC class I and, through association with p56lck, plays a role in T-cell development and activation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.25 µg)/test.
A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume (cell count: 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm; use with UV (355 nm) laser-equipped cytometers.

For panels containing multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (#SB-4401-42) or Brilliant Stain Buffer™ (#00-4409-75) to minimize non-specific interactions.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein functioning as a co-receptor for MHC class I molecules. It is expressed on T cells, NK cells, monocytes, and dendritic cells. CD8α is essential for MHC-I binding and contributes to T-cell activation. Upon antigen recognition, CD8 recruits Lck kinase, leading to cytokine production and cytotoxic T lymphocyte activation. Activated CTLs play a key role in pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
