Target Information
KLRB1 (Killer cell lectin like receptor subfamily B, member 1, CD161) is a type II membrane protein with an external C terminus. It is expressed on most NK cells and a subset of CD3⁺ T cells. KLRB1 functions as an inhibitory receptor on NK cells, influencing ceramide levels and kinase activation (AKT1/PKB, RPS6KA1/RSK1). It binds carbohydrate epitopes such as Gal-α(1,3)Gal and N-acetyllactosamine, and interacts with CLEC2D/LLT1 to modulate NK cell cytotoxicity and interferon-gamma secretion. Structurally, it includes extracellular C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
