The monoclonal antibody eBio4E5 (4E5) reacts with Ly-49D, an activating member of the Ly-49 family of NK cell receptors. These receptors form homodimers that recognize MHC Class I molecules.
Activating Ly-49 receptors (Ly-49D and Ly-49H) lack ITIM motifs in their cytoplasmic domains, distinguishing them from inhibitory Ly-49 members. Instead, they associate with DAP12, which contains an ITAM required for positive signaling.

Expression of Ly-49 family members is not linked, allowing multiple Ly-49 proteins to be expressed on the same NK cells.
Ly-49D expression is strain-dependent — found in C57BL/6, CB6F1, and SJL mice, but absent in BALB/c, DBA/2, AKR, and CBA/J strains.
In 129/J mice, eBio4E5 also recognizes Ly-49O, Ly-49V, and Ly-49R.
The antibody exhibits functional (activating) activity.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; use ≤ 0.5 µg/test (100 µL final volume). Cell numbers can range from 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Target Information

Ly-49D is an activating NK cell receptor recognizing MHC Class I molecules. It associates with DAP12 for ITAM-mediated signaling.
Expression varies by mouse strain and can overlap with other Ly-49 family members.
In 129/J mice, eBio4E5 cross-reacts with Ly-49O, Ly-49V, and Ly-49R.
