Product Specific Information
The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein. Two isoforms of Ki-67 exist (345 and 395 kDa) expressed in dividing cells. Ki-67 is present during active phases of the cell cycle (G1, S, G2, mitosis) but absent in resting cells (G0).
During interphase, Ki-67 localizes to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.
In tumor studies, Ki-67 expression is used as a marker to determine the fraction of proliferating cells.
This monoclonal antibody 20Raj1 also recognizes canine Ki-67.

Applications Tested:
- Tested by intracellular staining of normal human peripheral blood cells using Foxp3/Transcription Factor Buffers Set (Product #00-5523-00)
- Recommended staining protocol: BestProtocols® – Staining Intracellular Antigens for Flow Cytometry
- Use ≤0.125 µg/test (100 µL volume, 10⁵–10⁸ cells/test)
- For ICC, use ≤5 µg/mL
- Antibody should be titrated for optimal performance

Target Information
Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It contains a forkhead-associated (FHA) domain and associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Ki-67 shows cell cycle-dependent localization: perinucleolar (G1), nuclear matrix (G2), and chromosomal (M phase).
Used as a proliferation marker since absent in G0 cells and increases through mitosis.
Commonly used to determine the growth fraction in neoplasms; low Ki-67 index correlates with low-grade tumors.
Also used as a neuronal marker of cell cycling and proliferation.
