Product Specific Information
MA3-026 detects T Complex Polypeptide 1 (TCP-1) from human, canine, chicken, hamster, mouse, primate, rat, and yeast samples.
It has been successfully used in Western blot, immunofluorescence, and immunoprecipitation procedures.
- In Western blot: Detects a ~57 kDa band representing TCP-1 in mouse 3T3 cells.
- In immunofluorescence: Staining of TCP-1 in mouse germ line cells shows diffuse cytoplasmic staining.
The immunizing peptide corresponds to amino acid residues 306–556 from mouse TCP-1.
The epitope has been mapped to amino acids 465–469 (pentamer peptide AKLRA).
Target Information
T Complex Polypeptide 1 (TCP-1) is an approximately 60 kDa protein constitutively expressed in most eukaryotic cells and upregulated during spermatogenesis.
It is located in the cytosol as a subunit of a hetero-oligomeric chaperone complex involved in folding of actin and tubulin.
Chaperonins recognize and stabilize polypeptide intermediates during folding, assembly, and disassembly.
They share characteristics with Heat Shock Protein 70 (HSP70), including high abundance, stress induction, and ATPase activity.
The chaperonin family includes:
- Mitochondrial HSP60
- E. coli GroEL
- Plastid Rubisco-subunit binding protein
- Archaebacterial protein TF55
TCP-1 sequence shows about 40% identity to TF55, but minimal similarity to HSP60 and GroEL.
