Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein belonging to the TNFR family. CD40 is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and, together with IL-4, protects B cells from apoptosis and promotes proliferation. Interaction of CD40 with CD154 (gp39) is crucial for T-B cell crosstalk and immune regulation.

5C3 is used for activation of B cells in in vitro functional assays.

Applications Reported: Flow cytometric analysis and functional studies.
Applications Tested: Flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize by titration for best performance.

Storage and Handling: Use in a sterile environment.
Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).
Endotoxin Level: <0.001 ng/µg antibody (LAL assay).

Target Information

CD40 is a single-chain glycoprotein belonging to the TNF/NGF receptor superfamily and is homologous to CD30. It is expressed on all B cells except plasma cells, and also on epithelial cells, carcinomas, and dendritic cells. CD40 plays a key role in immune and inflammatory responses, including immunoglobulin class switching, memory B cell development, and germinal center formation.

AKNA transcription factor regulates CD40 expression, while TNFR2 mediates CD40 signal transduction. CD40-CD154 interaction is critical for amyloid-beta-induced microglial activation, implicated in Alzheimer’s disease. Two alternatively spliced CD40 isoforms are known. Associated diseases include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
