Product Specific Information

The monoclonal antibody L26 recognizes human CD20, a two-transmembrane spanning protein found on precursor and mature B lymphocytes. Expression is lost upon differentiation to plasma cells. CD20 levels are critical in evaluating malignant B lymphocytes, including Non-Hodgkin’s Lymphoma and various leukemias. CD20 plays a role in B cell differentiation and activation.

The L26 antibody recognizes an epitope in the cytoplasmic domain, requiring tissue permeabilization for staining. Unlike antibodies that target extracellular domains, L26’s epitope is not blocked by therapeutic antibodies such as Rituximab.

• Applications Tested: Tested by IHC on formalin-fixed paraffin-embedded human tonsil using IHC Antigen Retrieval Solution - Low pH (Product # 00-4955-58).

Fluorophore Information:
eFluor® 660 replaces Alexa Fluor® 647. It emits at 659 nm and is excited by a 633 nm red laser. Recommended usage ≤20 µg/mL; titration advised for optimal performance.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, and a subset of T lymphocytes and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 is associated with lipid rafts and participates in BCR-mediated signaling.

CD20 remains at the cell surface after antibody binding, making it a valuable target for antibody-mediated B-cell depletion therapies. Dysfunctions in CD20 are linked to Ms4a1-related common variable immune deficiency.
