Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events such as the establishment and maintenance of pregnancy.
PR exists as two major isoforms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), resulting from alternative translation initiation sites.
- PR-B: Transcriptionally active form, responsible for endometrial maintenance, pregnancy maintenance, and inhibition of ovulation.
- PR-A: Lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, PR undergoes conformational change, dimerizes, and binds to progesterone response elements (PRE), activating transcription.
A null mutation in the PGR gene results in pleiotropic reproductive abnormalities.
