Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress.
It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation.
p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest.
The structure includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation.
p53 plays roles in:
- Preventing proliferation of damaged cells
- Acting as an anticancer agent
- Linking to aging and oxidative stress
- Affecting learning and memory through LTP deficits
In normal cells, p53 levels are low, but in transformed cell lines, it is highly expressed and contributes to malignancy.
Mutations in TP53 cause loss of tumor suppressor activity and occur in many human cancers.
Germline mutations are also found in cancer-prone families such as Li-Fraumeni syndrome.
