Product Specific Information

1 µg/mL of MA5-45400 was sufficient for detection of HCN1 in 10 µg of rat brain lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody. Detects approximately 100 kDa. No cross-reactivity against HCN2.

Target Information

HCN1 (Hyperpolarization-activated cyclic nucleotide-gated channel 1) is part of a family of pacemaker channels activated by hyperpolarization and regulated by cyclic nucleotides. These channels contribute to spontaneous rhythmic activity in both the heart and brain. HCN1 and HCN2 are important for motor learning and neuronal integration by cerebellar Purkinje cells, as well as shaping autonomous neuronal activity and network oscillations. HCN1 is highly expressed in the cerebral cortex, hippocampus, cerebellum, and facial motor nucleus, while HCN2 is abundant in mamillary bodies, pontine nucleus, ventral cochlear nucleus, and nucleus of the trapezoid body. Regional variations in HCN channel expression contribute to differences in neuronal pacemaker activity across brain systems.
