Focal adhesions are areas within the plasma membrane that adhere tightly to the substrate. In vivo, they mediate cell adhesion to the extracellular matrix. Paxillin and vinculin are cytoskeletal focal adhesion proteins linking the actin network to the plasma membrane. Vinculin interacts with talin and α-actinin, forming a complex that connects actin filaments to integrins, playing a key role in cell adhesion and cytoskeletal organization.
Target Information
Vinculin (VCL) is a cytoskeletal protein associated with adherens-type junctions between cells and between cells and the extracellular matrix. It anchors F-actin to the membrane through interactions with talin, α-actinin, and integrins. Vinculin is essential for the structural integrity of adhesion plaques and cytoskeletal dynamics. Mutations in the VCL gene are associated with cardiomyopathy (dilated 1W or familial hypertrophic 15).
