The human ASH2L gene encodes a 628 amino acid protein (isoform 1, ASH2L1) containing a nuclear localization signal and a PHD finger motif, suggesting a role as a transcription regulator. The predicted molecular weight is approximately 68.7 kDa, with alternative isoforms at 60 kDa and 56.5 kDa.
ASH2L shares 60% homology with Drosophila ash2, which regulates early developmental gene expression, and 90% homology with mouse ASH2L. It is predominantly expressed in adult heart and testis, and fetal lung and liver, with low expression in adult lung, liver, kidney, prostate, and peripheral leukocytes. Expression varies in leukemia cell lines and is influenced by differentiation inducers such as phorbol ester and hemin, indicating a role in hematopoiesis and certain leukemia types. ASH2L is also suggested to interact with beta-tubulin and beta-catenin.
