Product Specific Information

This SPRCL5 monoclonal antibody reacts with mouse CD185 (CXCR5), also known as Burkitt lymphoma receptor 1 (BLR1). CXCR5 is a seven transmembrane G protein-coupled receptor expressed on B cells, CD4+ T cells, and memory T cells. It plays a critical role in lymphocyte trafficking and follicular helper T cell migration via CXCL13.

• Fixation: Staining should be done prior to fixation (does not recognize formaldehyde-fixed epitopes).
• Applications Reported: Flow cytometric analysis.
• Applications Tested: Flow cytometric analysis of mouse splenocytes at ≤0.5 µg/test (100 µL final volume).
  Cell number: 10⁵–10⁸ cells/test (empirically determined).
• Excitation/Emission: 488–561 nm / 607 nm; compatible with Blue, Green, and Yellow-Green lasers.
• Recommended Filter: 610/20 band pass (PE-Texas Red® equivalent).
• Light Sensitivity: Protect vial and stained samples from light (photo-induced oxidation sensitive).
• Fixation Storage: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (#00-8222) or 1-step Fix/Lyse Solution (#00-5333).
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CXCR5 (C-X-C Chemokine Receptor Type 5, CD185) is a G protein-coupled receptor expressed in Burkitt’s lymphoma and lymphatic tissues (lymph nodes, spleen). It mediates B cell migration and follicular organization. CXCR5 expression is also found in various human and animal tissues, including blood, bone marrow, brain, liver, and tonsil.

GPCRs, including CXCR5, share seven transmembrane helices and participate in complex signaling cascades. They are key pharmacological targets, with well-known drugs such as Zyprexa and Claritin developed against GPCRs.
