Product Specific Information

The AFKJS-9 monoclonal antibody reacts with mouse, human, and rhesus monkey ROR gamma (t) protein. ROR gamma belongs to the retinoic acid-related orphan receptor (ROR) family, including ROR alpha and ROR beta, which are ligand-dependent transcription factors involved in various physiological processes.

ROR gamma is expressed in liver, lung, muscle, heart, and kidney. An alternative transcript, ROR gamma (t), is expressed in lymphoid cells such as CD4+CD8+ thymocytes, Th17 cells, and lymphoid tissue inducer (LTi) cells. The ROR gamma (t) isoform differs by three amino acids at the N-terminus. Therefore, AFKJS-9 reacts with both ROR gamma and ROR gamma (t).

Applications Reported: Intracellular staining followed by flow cytometric analysis.

Applications Tested: Tested by intracellular staining and flow cytometric analysis of mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00). Recommended ≤1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize concentration for best results.

Fluorochrome Information:
PE-eFluor 610 is excited by 488–561 nm laser lines and emits at 607 nm. Use a 610/20 band pass filter (equivalent to PE-Texas Red). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light during storage and staining.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on signal quality. Clone-specific performance should be verified empirically.

Target Information

ROR gamma (NR1 family) influences thymopoiesis, bone metabolism, T-cell apoptosis, and lymphoid organogenesis. It promotes thymocyte survival by activating Bcl-x(L) expression and is required for lymph node and Peyer’s patch development. The thymus-specific isoform ROR gamma(t) inhibits Fas ligand expression and cytokine secretion in immature thymocytes.

ROR gamma binds as a monomer to response elements containing a single GGTCA motif with a 6 bp AT-rich sequence. Expression is found in mouse thymus, adipose, bone, skeletal muscle, liver, kidney, and human skeletal muscle. ESTs have been identified in various human tissues, both normal and cancerous.

ROR gamma(t) (RORgt) is an isoform of RORC.
