Product Specific Information

The CD28.2 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells.
CD28 serves as a ligand for CD80 (B7-1) and CD86 (B7-2), providing a potent co-stimulatory signal for T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and increases cytotoxicity of CD3-activated T cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells. Recommended use at 5 µL (1.0 µg) per test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test.

Fluorochrome: Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm and is intended for instruments with a 355 nm UV laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use:

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T-cell surface glycoprotein of the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and provides a crucial co-stimulatory signal by binding to CD80 and CD86 on antigen-presenting cells.
CD28 prevents T cells from entering an anergic or apoptotic state and is vital for proper immune activation. Expression patterns vary across thymocyte maturation stages.
Dysfunction of CD28 is associated with diseases such as mycosis fungoides and Sézary’s disease.
