Product Specific Information
The WM-53 monoclonal antibody reacts with human CD33, also known as GP67 and P67, a 67 kDa type I transmembrane glycoprotein that is a member of the Siglec (sialic acid-binding Ig superfamily lectin) family. It is highly specific to the hematopoietic compartment and is expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis on normal human peripheral blood cells.
Use at 5 µL (0.125 µg) per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test.

Target Information
CD33 is a transmembrane protein of the Siglec family, containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases. It is expressed on cells of myelomonocytic lineage and binds sialic acid residues on cell surfaces. CD33 is a therapeutic target for acute myeloid leukemia (AML) and serves as a marker for monocytes and myeloid precursors. It is not expressed on pluripotent stem cells.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
