Product Specific Information

The monoclonal antibody eBio440c recognizes Siglec-H, a protein exclusively found on plasmacytoid dendritic cells (pDCs) or type I IFN-producing cells (IPC) in naive mice. Mouse IPCs are typically PDCA+, CD11c+, CD11b-, B220+, and Ly-6C+, and respond rapidly to viral infection.

Siglec-H is a transmembrane protein of the Ig superfamily that, like CD33, binds sialic acid but lacks the cytoplasmic ITIM domain (immunoreceptor tyrosine-based inhibitory motif). Instead, Siglec-H associates with DAP12 for signal transduction. The eBio440c antibody has been shown to inhibit pDC function by reducing IFNα secretion in response to CpG stimulation.

Some mouse strains (e.g., SJL) exhibit higher percentages of pDCs compared to C57BL/6.

Target Information

Siglec-H is a pDC-specific transmembrane protein of the Ig superfamily associated with DAP12 for signaling. It binds sialic acid and plays a role in regulating pDC function, including IFNα secretion. Expression varies among mouse strains.
