Product Specific Information

Description:
The N418 monoclonal antibody reacts with mouse CD11c (integrin alpha X). CD11c non-covalently associates with beta 2 integrin to form the CD11c/CD18 heterodimer. It is expressed by dendritic cells, intestinal intraepithelial lymphocytes (IEL), and some activated T cells. CD11c/CD18 binds to CD54, iC3b, and fibrinogen, playing a role in leukocyte adhesion. N418 binds to CD11c on splenic dendritic cells in T-dependent areas of the mouse spleen and precipitates a 150, 90 kDa heterodimer.

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using mouse splenocytes. Recommended ≤0.25 µg/test (100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test). Optimize titration for best performance.

Fluorophore Information:
PE-eFluor® 610: Excitation 488–561 nm, Emission 607 nm.
Recommended filter: 610/20 band pass (PE-Texas Red® equivalent).
Ensure instrument compatibility.

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored up to 3 days at 4°C (dark) in:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)
  Clone-specific performance should be verified empirically.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD11, together with CD18, forms a heterodimeric adhesion molecule.

• CD11a: Expressed in B/T lymphocytes, macrophages, monocytes, neutrophils, basophils, eosinophils.
• CD11b: Found in monocytes, macrophages, NK cells, some B/T cells, granulocytes.
• CD11c: Expressed in monocytes, macrophages, NK cells, some granulocytes, and subsets of lymphocytes.
