Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimer or CD8αα homodimer. CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a critical role in T cell development and activation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test (defined as antibody amount per 100 µL cell sample). Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
PE-eFluor® 610 is excited by 488–561 nm laser lines and emits at 607 nm. Use a 610/20 band pass filter (PE-Texas Red® equivalent). Ensure instrument compatibility.

Handling Notes:

  • Light sensitivity: Protect vial and stained samples from light.
  • Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333).
  • Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family, essential for T cell signal transduction, differentiation, and activation. CD8 predominantly exists on T cells as CD8αβ heterodimers functioning as co-receptors for MHC-I molecules. CD8α chain mediates MHC-I binding and is also found on NK cells, monocytes, and dendritic cells as CD8αα homodimers. MHC-I ligation recruits Lck kinase, leading to lymphokine production and cytotoxic T cell activation. Activated CTLs are critical for pathogen and tumor clearance. Differentiation of naïve CD8⁺ T cells is enhanced by IL-2, IL-12, and TGF-β1.
