Product Specific Information
The eBio64DEC17 antibody reacts with human IL-17A and functions as a neutralizing antibody.
IL-17A is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
Human IL-17A exists as glycosylated 20–30 kDa homodimers produced by activated peripheral blood CD4+ T-cells. It enhances ICAM-1 expression in fibroblasts and stimulates epithelial, endothelial, or fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2.
IL-23-dependent, IL-17A-producing CD4+ T cells (Th17 cells) are a distinct subset of Th cells regulated by IFNγ and IL-4.
Intracellular staining by eBio64DEC17 identifies the same cell population as eBio64CAP17.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining and flow cytometry at 5 µL (0.125 µg)/test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test.
Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution for up to 3 days at 4°C in the dark.
Excitation: 488–561 nm; Emission: 775 nm; Laser: Blue, Green, Yellow-Green.

Target Information
Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine involved in inflammatory responses and elevated in autoimmune and inflammatory diseases.
It is a 32 kDa disulfide-linked homodimer of 136 amino acids, part of the IL-17A–F family, signaling through IL-17R/CDw217.
IL-17A enhances ICAM-1 expression and induces secretion of IL-6, IL-8, G-CSF, and PGE2, supporting hematopoietic progenitor proliferation and neutrophil maturation.
It activates NF-κB and MAPK pathways, increasing IL-6 and COX-2 expression and nitric oxide production.
