Product Specific Information

The eBio17B7 antibody reacts with mouse and rat IL-17A without recognizing IL-17F. IL-17A is a CD4+ T cell-derived cytokine involved in inflammatory responses and is elevated in rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
Human IL-17A exists as glycosylated 20–30 kD homodimers produced by activated CD4+ T-cells and promotes secretion of IL-6, IL-8, G-CSF, and PGE2 from epithelial and fibroblastic cells.

IL-23-dependent, IL-17A-producing CD4+ T cells (Th17 cells) represent a distinct subset from Th1 and Th2 cells. IFN-γ and IL-4 negatively regulate Th17 cell generation.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on restimulated Th17-polarized mouse splenocytes using Intracellular Fixation and Permeabilization Buffer Set (Product # 88-8824-00). Recommended ≤0.5 µg/test.
Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.
Compatible with Foxp3 buffer system (Product # 00-5523-00).

Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57).

Target Information

Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in autoimmune and inflammatory diseases.
IL-17A is a 32 kDa disulfide-linked homodimer of 136 amino acids, signaling through IL-17R/CDw217. It enhances ICAM-1 expression and stimulates secretion of IL-6, IL-8, G-CSF, and PGE2.
Mouse, rat, and human IL-17A can induce IL-6 secretion in mouse stromal cells, indicating cross-reactivity with the mouse receptor.
IL-17A regulates NF-κB and MAPK pathways, induces COX-2 expression, and enhances nitric oxide production.
