Product Specific Information
Immunohistochemistry (PFA fixed): 항체를 실온에서 30분간 반응시킵니다. 포르말린 고정 조직의 염색은 10 mM Tris, 1 mM EDTA, pH 9.0 용액에서 조직 절편을 95°C에서 45분간 가열 후 실온에서 20분간 냉각하는 과정을 필요로 합니다.
Target Information
GAD-65와 GAD-67은 group II decarboxylase 계열 단백질로, L-glutamic acid로부터 GABA(gamma-aminobutyric acid)를 생성하는 과정의 속도 제한 단계를 촉매합니다.
두 GAD 모두 뇌에서 발현되며, GAD-65는 신경 말단의 시냅스 소포막에 위치하고 GAD-67은 세포 전체에 분포합니다.
GAD-67은 기본적인 GABA 합성에 관여하며, 신경전달 시 GABA 요구량이 증가할 때 GAD-65가 일시적으로 활성화되어 GABA 생산을 보조합니다.
GAD-65의 결손은 GABA 신경전달에 장애를 초래할 수 있으며, GAD-67의 결손은 치명적입니다.
대체 스플라이싱으로 인해 GAD-67은 주요 형태(GAD-67)와 소수 형태(GAD-25)의 두 가지 isoform을 가집니다.
