• For Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Calponin is a thin filament-associated protein involved in the regulation of smooth muscle contraction. It binds to actin, calmodulin, troponin C, and tropomyosin, and is expressed in smooth muscle and tissues containing significant smooth muscle content.
Two isoforms exist (34 kDa and 29 kDa). The 29 kDa isoform is primarily expressed in urogenital tract muscle. Calponin is also expressed in myoepithelial cells of benign and malignant breast lesions.
It stains smooth muscle, myoepithelial cells, myofibroblasts, keratinocytes, and nerve fibers, aiding differentiation of breast collagenous spherulosis (positive) from adenoid cystic carcinoma. Adenoid cystic carcinoma in salivary gland tumors is calponin positive.
