• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Calponin is a thin filament-associated protein involved in the regulation and modulation of smooth muscle contraction. It can bind to actin, calmodulin, troponin C, and tropomyosin. Calponin is expressed in smooth muscle and tissues containing significant amounts of smooth muscle.

There are two isoforms of calponin with molecular weights of 34 kDa and 29 kDa. The 29 kDa form is primarily expressed in the urogenital tract. Calponin expression has also been observed in myoepithelial cells from benign and malignant breast lesions. It stains smooth muscle, myoepithelial cells, myofibroblasts, keratinocytes, and nerve fibers. It helps identify myoepithelial cells in breast lesions and differentiate breast collagenous spherulosis (positive) from adenoid cystic carcinoma. Adenoid cystic carcinoma in salivary gland tumors is calponin positive.
