This antibody detects endogenous levels of Nucleophosmin only when phosphorylated at Thr95.
Target Information
NPM1 (Nucleophosmin 1, also known as B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in:
- Ribosome assembly and transport
- Cytoplasmic/nuclear trafficking
- Regulation of DNA polymerase α activity
- Centrosome duplication
- Regulation of p53
NPM1 shuttles between the nucleus, cytoplasm, and nucleolus, acting as a chaperone for core histones. It binds nucleic acids, prevents protein aggregation, and assists in enzyme protection and refolding. NPM1 is associated with centrosome duplication and serves as a substrate for CDK2/cyclin E. It also interacts with tumor-associated fusion proteins (NPM-ALK, NPM-RAR, NPM-MLF1) and can sequester tumor suppressor proteins, such as ARF, within the nucleolus. Dysregulation of NPM1 is linked to diseases including acute myeloid leukemia and lymphomatoid papulosis.
