Mature human IL-1α was expressed in Pichia pastoris strain KM71H (GenBank Accession # P01583, amino acids 113–271).
Protein verification was performed by N-terminal sequencing, confirming the first 10 amino acids: SAPFSFLSNV.
Purity is >95% by SDS-PAGE analysis.

Activity:
The ED50 typically ranges from 7.9 to 13.5 pg/mL in proliferation assays using mouse D10S cells.

Use:
For in vitro applications. A dose-response assay should be performed to determine the optimal concentration for specific experiments.

Reconstitution and Dilution:

• Store lyophilized material at 4°C.
• Reconstitute in 0.5 mL of distilled or deionized water.
• Dilute in buffer containing carrier protein, aliquot, and store at or below –70°C.
• Avoid multiple freeze-thaw cycles.
• Suitable buffers include PBS with 1% BSA or neutral media containing <10% FCS.

Target Information

IL-1 alpha (Interleukin-1 alpha) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells.
It is encoded by the IL1A gene, located on chromosome 2q13, and signals through IL-1RI and IL-1RII receptors shared with IL-1β.

IL-1α plays a critical role in innate immunity by inducing proinflammatory cytokine production and acute-phase responses.
Its activity is regulated by IL-1 Receptor Antagonist (IL-1RA), which competitively inhibits receptor binding.
IL-1α influences NF-κB and MAPK signaling pathways, stimulates IL6 and COX-2 expression, and enhances nitric oxide (NO) production.
High IL-1α levels are associated with chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
