Target Information

SPHK2, along with SPHK1, catalyzes the phosphorylation of sphingosine to produce sphingosine-1-phosphate (S1P), a signaling molecule that acts through G-protein coupled receptors to regulate diverse cellular processes. Inhibitors of SPHKs are potential therapeutics for cancer treatment.
SPHK2 functions both intra- and extracellularly, regulating cell differentiation, calcium mobilization, and apoptosis.
S1P receptors (EDG family) interact with multiple G proteins (e.g., S1P1 with Gi; S1P2 and S1P3 with Gq, G13, and Gi) to control cell motility, survival, migration, and cell-cell interactions.
S1P also plays roles in cardiogenesis, vascular maturation, oocyte survival, immune cell trafficking, and neuronal and bone cell regulation.
S1P levels are modulated by Sphk1 and Sphk2 activity.
