Product Specific Information

This 1RNM44N monoclonal antibody recognizes mouse CD170 (Siglec F), a cell surface lectin of the Ig superfamily binding sialic acid-containing glycoconjugates. Siglec F is mainly expressed on eosinophils and alveolar macrophages and at lower levels on immature myelomonocytic cells. Mouse Siglec F is a functional ortholog of human Siglec 8 but is not expressed on mast cells.

• Recognizes a formaldehyde-fixed epitope
• Reported and tested for flow cytometric analysis
• Recommended use ≤ 1.0 µg/test (100 µL final volume)
• Cell count per test: 10⁵–10⁸ cells (empirical optimization required)

Fluorochrome Information:
Brilliant Violet™ 711 (BV711) emits at 713 nm and is suitable for cytometers with a violet (405 nm) laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use:

• Super Bright Complete Staining Buffer (Product # SB-4401-42)
• Brilliant Stain Buffer™ (Product # 00-4409-75)
  to minimize non-specific polymer interactions.

Light Sensitivity:
BV711 is sensitive to photo-induced oxidation — protect vial and samples from light.

Fixation Compatibility:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product # 00-8222-49)
• 1-step Fix/Lyse Solution (Product # 00-5333-54)

BV711 is not compatible with methanol-based fixation.

Target Information

SIGLEC5 is a putative adhesion molecule mediating sialic-acid-dependent binding to cells. It binds α2,3- and α2,6-linked sialic acid. The recognition site may be masked by cis interactions with sialic acids on the same cell surface. Expressed by monocytic/myeloid lineage cells, SIGLEC5 is found at high levels in peripheral blood leukocytes, spleen, and bone marrow, and at lower levels in lymph node, lung, appendix, placenta, pancreas, and thymus. Expressed by monocytes and neutrophils but absent from early leukemic myelomonocytic cell lines.
