Product Specific Information

The A7R34 monoclonal antibody reacts with mouse CD127, the high-affinity alpha subunit of the IL-7 receptor. CD127 is expressed in immature B cells, certain thymocyte subsets, and peripheral T cells at low levels. A7R34 inhibits IL-7 binding and is used to study IL-7’s role in lymphocyte development and activation. It also blocks binding of antibody SB/199 to mouse CD127.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize concentration empirically.

Fluorochrome Information:
Brilliant Violet™ 711 emits at 713 nm, suitable for instruments with a violet (405 nm) laser. Protect from light due to photo-induced oxidation sensitivity.

Staining Recommendations:
When using multiple polymer dyes (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use:

• Super Bright Complete Staining Buffer (SB-4401-42) or
• Brilliant Stain Buffer™ (00-4409-75)
  to minimize non-specific interactions.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (00-8222-49)
• 1-step Fix/Lyse Solution (00-5333-54)
  Brilliant Violet™ 711 is not compatible with methanol-based fixation.

Target Information

CD127 (Interleukin-7 receptor alpha, IL7Rα) is a glycoprotein regulating lymphopoiesis. It forms a heterodimeric receptor with the common gamma chain (γc), shared with IL-2 receptor. The alpha chain mediates specific signaling, promoting proliferation of precursor B cells, thymocytes, and mature T cells. CD127 dysfunction is associated with severe combined immunodeficiency (SCID) variants.
