Description:
The OX8 monoclonal antibody reacts with rat CD8 alpha, a member of the immunoglobulin superfamily. CD8α heterodimerizes with CD8β to form CD8 antigen expressed on CD4+CD8+ thymocytes, MHC class I-restricted T cells, and some macrophages. CD8 binds MHC class I on antigen-presenting cells and participates in T-cell receptor signaling through Lck kinase association.

Applications Reported:
This OX8 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended ≤0.5 µg per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Fluorochrome Information:
eFluor 450 is an alternative to Pacific Blue. Emits at 446 nm, excited at 405 nm (violet laser). Ensure instrument compatibility.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family involved in signal transduction and T cell differentiation. CD8 is mainly expressed on T cells as a CD8α/CD8β heterodimer acting as a co-receptor for MHC class I molecules. CD8α also forms homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Interaction with MHC-I/peptide complexes recruits Lck kinase, leading to lymphokine production and cytotoxic T lymphocyte activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
