Product Specific Information
The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as either a heterodimer (CD8αβ) or homodimer (CD8αα).
- CD8αβ is expressed on most thymocytes and mature αβ TCR T cells.
- CD8αα is expressed on γδ TCR T cells, intestinal intraepithelial lymphocytes (IELs), and dendritic cells.
CD8 binds to MHC class I and, via association with p56lck, plays a key role in T cell development and activation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes; optimal use ≤ 0.25 µg/test (100 µL final volume).
Fluorophore: eFluor® 450 (alternative to Pacific Blue®).
Excitation: 405 nm; Emission: 445 nm; Laser: Violet.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family.
- Functions as a co-receptor with TCR for MHC class I molecules.
- Expressed on T cells, NK cells, monocytes, and dendritic cells.
- CD8α chain is essential for MHC-I binding.
- Engagement with MHC-I/peptide complexes triggers Lck activation, leading to cytokine production, cytotoxic T lymphocyte (CTL) activation, and immune response regulation.
- Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
