Product Specific Information
The W6/32 monoclonal antibody reacts with human major histocompatibility complex (MHC) class I, HLA-A, B, and C. These antigens, associated with beta2-microglobulin, are expressed by all nucleated cells and play a key role in cell-mediated immune response and tumor surveillance.
W6/32 mAb recognizes a non-polymorphic epitope shared among HLA-A, B, and C loci and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity is observed in baboon, rhesus, and cynomolgus monkey.
Applications Reported: Flow cytometric analysis, immunoprecipitation, and immunohistology staining of frozen tissue sections.
Applications Tested: Flow cytometric analysis of normal human peripheral blood cells (≤1 µg/test). A test is defined as the amount of antibody that stains a cell sample in a 100 µL final volume (typically 10⁵–10⁸ cells/test). Antibody titration recommended for optimal performance.

Target Information
HLA-class I MHC antigens are intrinsic membrane glycoproteins expressed on nucleated cells, noncovalently associated with beta2-microglobulin. They present foreign determinants for immune recognition by cytotoxic T cells, essential for antiviral and antitumor defense.
Human HLA-class I antigens include HLA-A, HLA-B, and HLA-C molecules.
HLA and MHC antibodies are critical in Immunopeptidomics, enabling identification and characterization of neoantigens via HPLC-MS/MS.
