Target Information
DNA-directed RNA polymerase II subunit RPB1 (POLR1A) is a DNA-dependent RNA polymerase that catalyzes the transcription of DNA into RNA using ribonucleoside triphosphates as substrates. POLR1A is the largest subunit and a catalytic component of RNA polymerase II, which synthesizes mRNA precursors and non-coding RNAs. It forms the polymerase active center with the second largest subunit.
Pol II is the central component of the basal RNA polymerase II transcription machinery, composed of mobile elements that move relative to each other. RPB1 contributes to the core element with the central cleft, clamp, and jaws that interact with the DNA template.
During transcription initiation, the promoter DNA strand is positioned within the active site cleft. A bridging helix from RPB1 promotes translocation of Pol II by switching conformations, facilitating nucleotide addition. The phosphorylation status of the C-terminal domain (CTD) regulates transcription initiation, elongation, termination, and mRNA processing.
