Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and some dendritic cells. CD14 binds LPS-LBP complexes with high affinity on monocyte surfaces.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.6 µg) per test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures engineered for optimized fluorescence with minimal spillover.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 binds LPS via LBP and plays roles in immune response, apoptosis recognition, and inflammatory processes.
Dysfunction of CD14 has been associated with infections such as Mycobacterium chelonae and conditions like croup.
Alternative splicing produces multiple transcript variants encoding the same isoform.
