Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and some dendritic cells. CD14 binds LPS-LBP complexes with high affinity.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using human peripheral blood cells. Recommended usage: 5 µL (0.6 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) required for optimal staining. Add 5 µL per sample to antibody cocktails before combining with cells.

Technology:
NovaFluor conjugates utilize Phiton™ technology based on engineered nucleic acid dye structures for optimized fluorescence with reduced spillover.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 participates in immune responses including recognition of LPS, apoptotic cells, and modulation of inflammatory processes. Associated diseases include mycobacterium chelonae infection and croup.
