Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and some dendritic cells. CD14 binds LPS-LBP complexes with high affinity on monocyte surfaces.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.6 µg) per 100 µL cell sample (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL of buffer per sample before mixing with cells.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It serves as a multifunctional receptor for lipopolysaccharides (LPS) and participates in immune recognition and inflammatory processes. Soluble CD14 (sCD14) binds LPS in serum and plays a role in immune modulation. CD14 dysfunction is associated with infections such as Mycobacterium chelonae and respiratory diseases like croup.
