Target Information

IkB-alpha is a 40 kDa protein that functions to inhibit NF-κB activity through protein-protein interaction with NF-κB dimers in the cytosol. This interaction masks the nuclear localization sequence of NF-κB, preventing its translocation to the nucleus.
Various stimuli such as TNF-α, IL-1β, chemokines, PMA, growth factors, LPS, UV irradiation, viral infection, and other stresses can activate NF-κB by promoting degradation of IkB-alpha.

In humans, the gene is located on chromosome 14q. Activation of NF-κB requires phosphorylation of IkB on serine residues (Ser32 and Ser36), leading to its degradation. IkB kinase alpha (IKKα), also known as CHUK, phosphorylates IkB-alpha at these sites, which is essential for NF-κB activation in response to proinflammatory cytokines.
IKKα activity is regulated by NF-κB inducing kinase (NIK), a central regulator of NF-κB activation. The functional IKK complex includes IKKα, IKKβ, and IKKγ, all contributing to IkB phosphorylation.
