The protein encoded by this gene is an unusual orphan receptor that contains a putative ligand-binding domain but lacks a conventional DNA-binding domain.
It belongs to the nuclear hormone receptor family, a group of transcription factors regulated by small hydrophobic hormones.
Some members of this family, including NR0B2, are referred to as orphan nuclear receptors as they lack known ligands.
This protein interacts with retinoid and thyroid hormone receptors to inhibit ligand-dependent transcriptional activation, and also interacts with estrogen receptors, leading to inhibition of their function.
Studies suggest that NR0B2 represses nuclear hormone receptor-mediated transactivation via two mechanisms: competition with coactivators and direct transcriptional repression.
