Target Information
Metadherin (Metastasis adhesion protein), also known as MTDH or LYRIC, is a novel protein that localizes with tight junction proteins ZO-1 and occludin in polarized epithelial cells. It is recruited during tight junction maturation rather than serving as a structural component.
Metadherin is overexpressed in breast cancer tissue and xenografts, with significantly lower levels in normal breast tissue. It binds to lung vasculature — a common site of breast cancer metastasis — via a C-terminal segment in its extracellular domain. Blocking this domain with antibodies or inhibiting metadherin using siRNA has been reported to suppress breast cancer metastasis.
