Product Specific Information
This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios. Helios is a zinc-finger transcription factor related to Ikaros, expressed mainly in regulatory T cells but not in mature B cells, dendritic cells, or myeloid cells. It can form homo- and heterodimers with other Ikaros family members to regulate transcription. Some isoforms act as dominant-negative variants and are upregulated in lymphoid malignancies such as T-acute lymphoblastic leukemia.
22F6 has been reported to cross-react with rat Helios.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested using mouse lymph node cells with Foxp3/Transcription Factor Buffer Set (Product # 00-5523-00).
Recommended staining protocol: BestProtocols® - Staining Intracellular Antigens for Flow Cytometry.
Usage: 5 µL (0.00375 µg) per test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.
PerCP-eFluor® 710 emits at 710 nm and is excited by a 488 nm blue laser.
Recommended filter: 710/50 bandpass (alternative: 695/40 bandpass).
Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Helios is a member of the Ikaros family of zinc-finger proteins (Ikaros, Aiolos, Helios), which are hematopoietic-specific transcription factors regulating lymphocyte development. Helios forms homo- or heterodimers with other family members and functions primarily in early hematopoietic development. Multiple transcript variants exist, though not all have confirmed biological validity.
