Target Information
KIT (c-KIT)은 MGF(mast cell growth factor, stem cell factor)의 3형 수용체로 작용하는 원암유전자(proto-oncogene)입니다.
이 유전자는 고양이 육종 바이러스의 v-kit과 상동 관계를 가지며, 다양한 조혈세포 및 비조혈세포의 성장과 활성화를 조절합니다.
KIT의 돌연변이는 위장관 기질종양(GIST), 비만세포 질환, 급성 골수성 백혈병(AML), piebaldism 등과 관련이 있습니다.
특히 활성 루프 변이 D816V는 비정상적인 KIT 수용체 활성화를 유도하여 종양성 비만세포 성장의 주요 치료 타겟으로 연구되고 있습니다.
c-Kit은 조혈줄기세포, 생식세포, 비만세포, 위장관의 Cajal 세포 등에 발현되며, SCF(stem cell factor) 결합 시 이합체화되어 자가 인산화 및 다양한 신호전달 경로(PI3K, PLC-γ, JAK/STAT 등)를 활성화합니다.
이는 조혈, 줄기세포 유지, 생식세포 형성에 필수적인 역할을 합니다.
