Preparation:
Produced from sera of goats immunized with highly pure Recombinant Murine EGF. Anti-Murine EGF-specific antibody was purified by affinity chromatography employing an immobilized Murine EGF matrix.
Sandwich ELISA:
To detect Murine EGF by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Murine EGF (500-P174GBt) as a detection antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Murine EGF.
Western Blot:
To detect Murine EGF by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. Used in conjunction with compatible secondary reagents, the detection limit for Recombinant Murine EGF is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.
Target Information
EGF (Epidermal Growth Factor) binds to the EGF receptor (EGFR), a 170 kDa protein kinase. Activation of EGFR initiates diverse cellular pathways in response to environmental stimuli. Upon EGF binding, EGFR forms homo- or heterodimers with other family members, triggering distinct signaling cascades via recruitment of SH2-containing effector proteins.
EGF is widely expressed in kidney, cerebrum, prostate, and salivary glands. It acts as a potent mitogenic factor, activating downstream signaling pathways such as:
- RAS-RAF-MEK-ERK
- PI3K-AKT
- PLCγ-PKC
- STAT
EGF may also influence NF-κB signaling. Mutations in the EGF gene are associated with hypomagnesemia type 4, and dysregulation is linked to cancer progression.
