Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris Buffer with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily.
It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR.
TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It typically exists as multimers (dimers, trimers, or pentamers) but appears as a single 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies in mice suggest TNF alpha has neuroprotective functions and induces tumor necrosis in vivo.
It plays key roles in immune responses to bacterial, viral, parasitic, and fungal infections, and contributes to tumor cell cytolysis and cytostasis, often synergistically with interferon-gamma.
TNF alpha is implicated in autoimmune diseases, insulin resistance, and cancer.
