Target Information
Humanin is a novel 24 amino acid polypeptide identified in Alzheimer’s disease-affected brain. It is 99% homologous to mitochondrial 16s ribosomal RNA and functions as a neuroprotective and cytoprotective factor. Humanin inhibits neuronal cell death induced by beta-amyloids and Alzheimer’s disease-causing FAD genes by competitively inhibiting FPRL1 receptor access to beta-amyloids. It acts as an anti-apoptotic factor by binding to Bcl-2/Bax family proteins (Bax, Bak, Bid, BimEL) and inhibiting their pro-apoptotic activity. Humanin activates the JAK/STAT signaling pathway, is regulated by TRIM11, and interacts with IGFBP-3 to block IGFBP-3–induced cell death in glioblastoma cells. It is also expressed in colon and testis.
