Target Information

The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). The His tag is commonly expressed at either the N- or C-terminal region of recombinant proteins, allowing isolation or purification via immobilized metal affinity chromatography (IMAC).

Epitope tagging is a technique in which a known epitope is fused to a recombinant protein using genetic engineering. This enables detection of proteins for which no specific antibody is available.

His-tagged proteins containing a stretch of histidine residues at the carboxyl or amino terminus are purified by metal chelate chromatography due to the metal-binding properties of histidine clusters. The affinity of the histidine-tag motif to Ni²⁺ ions allows selective purification to homogeneity using Ni²⁺-NTA adsorbents.

His tag-specific antibodies are used for detection or co-immunoprecipitation of tagged proteins.
