Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
It is commonly expressed at either the N- or C-terminal regions of recombinant proteins to enable isolation or purification by immobilized metal affinity chromatography (IMAC).
Epitope tagging allows detection of recombinant proteins for which no specific antibody is available. His-tagged proteins can be purified via metal chelate chromatography due to the metal-binding properties of histidine clusters.
For example, the strong and selective affinity of the His-tag motif to Ni²⁺ enables purification to homogeneity using Ni²⁺-NTA adsorbent.
His tag-specific antibodies facilitate detection and co-immunoprecipitation of tagged proteins.
