Product Specific Information

This antibody recognizes the extracellular part of CD30 (Ki-1 antigen), a 105 kDa single-chain glycoprotein expressed on Hodgkin’s and Reed-Sternberg cells. It is also found in Burkitt’s lymphomas, virus-infected T and B lymphocytes, and on normal B and T lymphocytes after activation, particularly on Th2-type cytokine-producing T lymphocytes and CD4+/CD8+ T lymphocytes co-expressing CD45RO and IL4 receptor.


Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein of the TNF receptor superfamily. It was originally identified as a cell surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. The ligand for CD30 is CD30L (CD153). The CD30–CD30L interaction mediates effects such as cell proliferation, activation, differentiation, and apoptosis. CD30 plays a critical role in Hodgkin’s disease and other CD30+ lymphomas and acts as a costimulatory molecule in thymic negative selection.

CD30 is expressed on Hodgkin’s and Reed-Sternberg cells, some non-Hodgkin’s lymphomas (including Burkitt’s lymphomas), virus-infected T and B cells, and activated normal T and B cells. In T cells, CD30 is expressed on a subset producing Th2-type cytokines and on CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor. Soluble CD30 (sCD30) serves as a marker of Th2 immune response. TRAF2 and TRAF5 interact with CD30 to activate NF-κB signaling. CD30 positively regulates apoptosis and limits autoreactive CD8 effector T cell proliferation, protecting against autoimmunity. Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
