Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen hormone, induces a conformational change allowing dimerization and binding to estrogen response elements (ERE) in DNA. Upon binding, ER regulates gene transcription by recruiting coactivator or corepressor proteins.
There are two forms of the estrogen receptor, alpha and beta, encoded by separate genes (ESR1 and ESR2). Alternative RNA splicing produces at least four alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are expressed in various tissues and are essential for sexual development, reproductive function, and bone physiology. They are also implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
