Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor acts as a ligand-activated transcription factor that, upon binding to estrogen, undergoes conformational changes enabling dimerization and DNA binding to estrogen response elements (ERE). This interaction regulates gene transcription through coactivator or corepressor recruitment.
There are two receptor forms, alpha (ESR1) and beta (ESR2), encoded by separate genes. Alternative RNA splicing produces at least four ESR1 isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are expressed in various tissues and are vital for sexual development, reproductive function, and bone health. They are implicated in diseases such as breast and endometrial cancer, and osteoporosis.
